Our scientists explain the components of the MSI-plus assay
The MSI-plus assay kit is a 96-well plate where each well contains a mix of 21 primer pairs including:
- 14 primer pairs for the 14 mononucleotide repeats used in MSI analysis
- 7 primer pairs for 22 mutation hotspots in BRAF, KRAS, and NRAS.
The primers incorporate Illumina adapter sequences, with a unique combination of i5 and i7 sample index sequences in each well, allowing for up to 96 distinct samples to be sequenced per batch.
The additional materials named in the protocol are all standard, widely-available laboratory reagents and consumables, such as the polymerase and Illumina sequencing kits; these are purchased separately.
The MSI-plus assay analysis pipeline uses free, open-source software, which is provided as a package by our team with support to set it up on your computer or integrate it into your bioinformatics infrastructure.
